Methods include Edman degradation, which sequentially removes and identifies N-terminal residues, and tandem mass spectrometry (MS/MS), which fragments peptides and deduces sequences from fragmentation patterns. Sequence confirmation is a critical quality control step in peptide manufacturing, ensuring that the synthesized product matches the intended target sequence and that no deletion, insertion, or substitution errors occurred during solid-phase peptide synthesis.
