Peptide Reconstitution Protocol
Step-by-step protocol for reconstituting lyophilized peptides with bacteriostatic water, including proper technique, concentration calculations, and storage guidelines.
Materials Needed
- Lyophilized peptide vial
- Bacteriostatic water (0.9% benzyl alcohol)
- Sterile syringe (1 mL insulin syringe recommended)
- Alcohol swabs (70% isopropyl)
- Clean workspace or laminar flow hood
- Powder-free nitrile gloves

Prepare Your Workspace
Clean your work surface thoroughly with 70% isopropyl alcohol. Put on powder-free nitrile gloves. Gather all materials before beginning. If available, work within a laminar flow hood for optimal sterility.

Allow Peptide to Reach Room Temperature
Remove the lyophilized peptide vial from cold storage and allow it to equilibrate to room temperature for 10-15 minutes. Do not attempt to heat the vial. Opening a cold vial can introduce moisture condensation into the powder.

Swab Vial Tops
Remove the protective caps from both the peptide vial and the bacteriostatic water vial. Swab the rubber stoppers of both vials with separate alcohol swabs. Allow the alcohol to air dry completely (approximately 30 seconds).

Do not blow on the stoppers to dry them — this introduces bacteria
Draw Bacteriostatic Water
Using a sterile syringe, draw the desired volume of bacteriostatic water. Use the reconstitution calculator below to determine the correct volume based on your target concentration. Common reconstitution volumes are 1 mL or 2 mL.

- • Pull back on the syringe plunger slightly past your target volume, then push forward to the desired volume to remove air bubbles

Add Water to Peptide Vial
Insert the needle through the rubber stopper of the peptide vial at a slight angle. Slowly inject the bacteriostatic water down the inside wall of the vial — do NOT spray directly onto the lyophilized powder. The stream should gently run down the glass interior wall and contact the powder at the bottom.

- • Aim the needle tip toward the glass wall, not the center of the vial
- • A slow, steady stream preserves peptide integrity better than rapid injection
Allow Dissolution
After adding the water, set the vial on a flat surface and allow the peptide to dissolve naturally. Most peptides will dissolve within 2-5 minutes. You may gently swirl the vial with a rolling motion between your palms, but never shake vigorously or vortex.

Verify Solution Clarity
Hold the vial up to a light source and inspect the solution. A properly reconstituted peptide should be clear and free of visible particles or cloudiness. If particulate matter is visible, do not use the solution.

Label and Store
Label the vial with the peptide name, concentration, reconstitution date, and your initials. Store the reconstituted peptide at 2-8°C (standard refrigerator). Most reconstituted peptides remain stable for 21-30 days when stored properly.
